岩白菜可培养内生细菌多样性分析 被引量:4

摘要 岩白菜[Bergenia purpurascens(Hook.f.et Thoms.)Engl.]是虎耳草科岩白菜属(Bergenia)具有很高观赏价值的一种多年生药用草本植物。分离并鉴定岩白菜的内生细菌,以揭示其可培养内生细菌的多样性。本研究以栽培和野生岩白菜为研究对象,将岩白菜组织块进行培养,采用菌株形态和分子生物学相结合的方法对分离的菌株进行了研究。共分离得到119株内生细菌,栽培品和野生品的根状茎和叶片中分离到的内生细菌分别为31,26,33,29株。对菌株的16S rDNA基因片段序列测定,结合其形态学特征分析,结果表明:芽孢杆菌属(Bacillus)和假单孢属(Pseudomonas)为岩白菜内生细菌的优势属,分别占总数的44%和12%。在内生细菌多样性程度上,野生品比栽培品高,根状茎比叶片高。 Bergenin [ Bergenia purpuras cens ( Hook. f. et Thorns. ) Engl. ] is a perennial medici- nal herbs with high ornamental value of Saxifragaceae Bergenia. Endophytic bacteria in rhizomes and leaves of wild and cultivated B. purpurascens were isolated and identified to reveal the diversity of en- dophytic bacteria. We took cultivated and wild Bergenia as the object of research, culturing Bergenia explant, and used the method combining with Morphology of strain and molecular biology to study the isolate strains. A total of 119 endophytic bacteria strains were isolated. The numbers of bacteria strains from rhizomes and leaves of cultivated and wild B. purpurascens were 31, 26, 33, 29, respectively. Based on morphology and the sequence analysis of 16S rDNA gene, Bacillus and Pseudomonas were dominant endophytic bacteria in B. purpurascens, accounting for 44% and 12% of total endophytic bacteria. Wild sample was higher than cultivated sample and rhizome sample was higher than leaf sam- ple at the diversity degree of endophytic bacteria.
