Inhibitor of nuclear factor kappa-B(IκB), an important member of nuclear factor kappa B(NF-κB) signaling pathway, is an essential adaptor in the regulation of NF-κB transcriptional activity. It is currently accepted that IκB plays an essential role in innate immunity and adaptive immunity, infectious diseases, autoimmune diseases, cancers and genetic diseases. Here we will review the structure, fundamental function of IκB and its role in signal transduction. Furthermore, we will also illustrate the relationship between IκB and the autoimmune diseases or infectious diseases.
