摘要
目的探讨羟基红花黄色素A(HYSA)抗四氯化碳所致肝纤维化的作用及对TGF-β1/CTGF表达的影响,为探索HYSA抗肝纤维化治疗的应用提供实验依据。方法将SD大鼠36只随机分为对照组、模型组和HYSA组(每组12只),以四氯化碳制备大鼠肝纤维化模型,HYSA组给予10 mg/kg HYSA治疗,8周后通过HE染色和Masson染色对肝纤维化程度进行评分;免疫组织化学观察TGF-β1、CTGF表达。结果 HYSA组与模型组相比,在纤维沉积面积[(2.168±0.160)%vs.(3.984±0.398)%]、纤维化评分(9.250±4.126 vs.19.333±3.448)、TGF-β1表达(0.184±0.018vs.0.287±0.019)及CTGF表达(0.082±0.011 vs.0.129±0.015)差异均具有统计学意义(P<0.05)。结论 HYSA可明显减轻四氯化碳所致的肝纤维化的程度,HYSA可能通过抑制TGF-β1和CTGF的表达发挥抗肝纤维化的作用。
Objective To investigate the therapeutic effects of HYSA on hepatic fibrosis in rats and the mechanisms of TGF-β1/CTGF pathway involved in this process. Methods The rats were divided into three groups: the normal control group, model group and HYSA group, the model of hepatic fibrosis were induced by carbon tetrachloride. The model group and experimental group were treated with saline and HYSA respectively for 8 weeks, to determine the pathological changes and grades of hepatic fibrosis, the obtained slides of liver tissue were stained by HE and Masson. The expression of TGF-β1 and CTGF were detected by means of immunohistochemistry. Results In the HYSA group, the area of fiber deposition(2.168±0.160)%, the hepatic fibrosis scores(9.250±4.126), the expression of TGF-β1 protein(0.184±0.018) and CTGF protein(0.082±0.011) were lower than those of the model group [(3.984±0.398)%,19.333±3.448, 0.287±0.019, 0.129±0.015], the differences were significant(P0.05). Conclusion HYSA significantly decreased the liver fibrosis level that caused by CCl4 HYSA may reduce rat hepatic fibrosis by TGF-β1/CTGF pathway.
