维生素A纳米微乳的吸收动力学研究 被引量:6

摘要 以海赛克斯蛋鸡为试验动物,采用高效液相色谱法(HPLC)检测血清视黄醇在口服不同剂型维生素A(VA)后的动态变化。所得数据用PKSolver 2.0软件进行药代动力学相关参数分析,结果表明:脂溶型VA的血药峰浓度(Cmax)为(0.29±0.02)μmol/mL,达峰时间(Tmax)为(2.47±0.10)h,药时曲线下面积(AUC0~t)为(1.44±0.24)μmol/mL·h,吸收半衰期(t1/2α)为(1.68±0.01)h;水溶型VA的Tmax和t1/2α分别为(1.44±0.15)h和(0.92±0.01)h,均显著低于脂溶型VA(P〈0.05),其余各参数与脂溶型VA相比无显著差异;纳米型VA的Tmax和t1/2α分别为(1.19±0.02)h和(0.83±0.03)h,均极显著低于脂溶型VA(P〈0.01),纳米型VA的Cmax为(0.38±0.004)μmol/mL,极显著高于脂溶型VA(P〈0.01),AUC0~t为(1.99±0.29)μmol/mL·h,显著高于脂溶型VA(P〈0.05)。以脂溶型VA为参比剂型,纳米微乳型VA的相对生物利用度为138.19%(P〈0.05)。综上,纳米微乳型VA在吸收速率、吸收峰值以及生物利用度方面优于脂溶型VA。 Dynamic change of the retinol from plasma samples of Hexics Brown hens were detected by HPLCafter oral administration with different forms of vitamin A,including lipo-soluble(lsVA),water-soluble(wsVA)andnano-emulsion vitamin A(neVA). The data were analyzed to obtain the pharmacokinetics parameters via the software ofPKSolver2.0.The results showed as following:as for lsVA,the peak concentration(Cmax)was(0.29±0.002)μmol/mL,timeto peak(Tmax) was(2.47±0.10)h,the area under the concentration-time curve(AUC0~t)was(1.44±0.24)μmol/mL·hand the absorption half life time(t1/2α)was(1.68±0.01)h. While the Tmax and t1/2αof wsVAwere(1.44±0.15)h and(0.92±0.01)h,respectively,which were significantly lower than those of lsVA. The Tmax and t1/2αof neVAwere(1.19±0.02)h and(0.83±0.03)h,respectively,which were significantly lower than those of lsVA,while the Cmax was(0.38±0.004)μmol/mL,which was significantly higher than that of lsVA.The AUC0~tof neVAwas(1.99±0.29)μmol/mL·h,which was significantly higher than that of lsVA. Compared with lsVA,the relative bioavailability of neVAwas 138.19%(P0.05). To sum up,neVAwas superior to lsVAin absorption rate,peak absorption and bioavailability.
