摘要
目的探讨葡萄籽提取物(GSE)对人骨肉瘤U2OS细胞的抑制作用及其作用机制。方法选用不同浓度的GSE作用于骨肉瘤U2OS细胞,MTT法检测其生长抑制作用;流式细胞仪检测细胞凋亡率;Transwell迁移实验检测细胞迁移能力;实时RT-PCR和Western blot检测CDC20mRNA和蛋白的表达水平。结果 MTT结果显示GSE对人骨肉瘤U2OS细胞的生长抑制作用呈浓度依赖性;流式细胞术显示GSE具有诱导U2OS细胞凋亡的作用;Transwell实验显示GSE可以抑制U2OS细胞的迁移作用;RT-PCR和Western blot结果显示GSE可以下调CDC20mRNA和蛋白的表达水平。结论 GSE具有抑制骨肉瘤U2OS细胞生长及迁移、诱导细胞凋亡的作用。GSE对骨肉瘤细胞的抑制作用可能与下调CDC20表达有关。
Objective To explore the inhibitory effects of grape seed extract(GSE)on human osteosarcoma U2OS cells and the possible mechanism.Methods After treatment of human osteosarcoma U2OS cells with GSE of difference concentrations, the cell growth inhibition was detected by MTT assay,the cell apoptosis by flow cytometry,the cell migration by Transwell assay,the mRNA and protein expression of CDC20by real-time RT-PCR and Western blot,respectively.Results MTT assay showed that GSE inhibited the growth of U2OS cells in a dose-dependent manner.Flow cytometry showed that GSE could induce cell apoptosis.Transwell assay showed that GSE could significantly inhibit the migration of U2OS cells.RT-PCR and Western blot results displayed that GSE could down-regulate the expression of CDC20mRNA and protein.Conclusion GSE can inhibit the proliferation and migration of human osteosarcoma U2OS cells and induce cell apoptosis.The effects of GSE are associated with the down-regulation of CDC20.
