BACE1及其相关蛋白在动脉粥样硬化性血管病中的表达

The expression of BACE1 and its associated proteins in atherosclerotic vascular disease

摘要 目的 观察动脉粥样硬化模型大鼠血管组织中淀粉样前体蛋白β-位分解酶1 (β-site APP-cleaving enzyme 1,BACEl)、β-淀粉样前体蛋白(β-amyloid precursor protein,APP)、β-淀粉样蛋白(β-amyloid peptide,Aβ)的表达,以探讨其在动脉粥样硬化疾病中的可能作用.方法 12只雄性SD大鼠随机分为2组,正常组(n=6)给予普通鼠饲料喂养,模型组(n=6)给予高脂高胆固醇饲料造模.3个月后腹主动脉取血测定两组大鼠血脂含量;HE染色观察胸主动脉血管壁形态学变化;免疫组织化学法观察大鼠胸主动脉壁组织中的BACE1、Aβ表达差异;RT-PCR法检测颈总动脉壁组织中BACE1、APP的mRNA表达.结果 模型组和对照组相比,各血清学指标均明显升高(P< 0.05或P<0.01);模型组动脉粥样硬化病变更为显著,动脉壁内BACE1、Aβ蛋白表达增加;与对照组相比,模型组BACE1、APP mRNA表达均升高[(1.009±0.16) VS (2.443±0.34),P< 0.01;(1.030±0.32) VS(1.818±0.21),P< 0.05].结论 BACE1及其相关蛋白在动脉粥样硬化大鼠血管组织表达均增高,其可能参与了动脉粥样硬化病的发病过程。 Objective To investigate expression of BACE1, APP, and A β in vascular tissue of atheroselerosis rats, and study the possible action of atherosclerosis. Methods 12 male SD rats were randomly divided into two experimental groups, the control group which was assigned to receive normal diet (n=6), and the model group which was given high fat and cholesterol diet (n=6) for a total of 3 months. Biochemical assay for blood cholesterol and HE staining for morphology changes of thoracic artery were observed, as well as Immunohistochemistry for BACE1 and A β of rat thoracic artery and RT-PCR for BACE1, APP of rat carotid artery. Results The levels of serologic indexes in model group were obviously higher than those in control group (P〈0.05 or P〈0.01). Atherosclerotic lesions and the expression of BACE1, A β in vascular increased significantly in model group. Comparing with the control group, the BACE1, APP mRNA expression were increased in all atherosclerotic model group (1.009±0.16 vs. 2.443±0.34, P〈 0.01; 1.030±0.32 vs. 1.818±0.21, P〈 0.05). Conclusion The expression of BACE1 and its associated proteins increased in atherosclerosis rats It may participate in the pathogenesis of atherosclerotic disease.
