摘要
目的研究姜黄素对香烟烟雾提取物刺激巨噬细胞株RAW264.7所致炎症反应的抑制作用。方法将RAW264.7巨噬细胞株培养于含不同浓度香烟烟雾提取物或姜黄素的培养基,MTT法筛选合适的实验浓度;组蛋白去乙酰化酶抑制剂曲古抑菌素A或香烟烟雾提取物分别刺激RAW264.7细胞株,体外模拟COPD发病的炎症反应过程;应用姜黄素和/或地塞米松处理被曲古抑菌素A或香烟烟雾提取物活化的RAW264.7细胞株;ELISA法测定细胞培养上清中炎症因子TNF-α、IL-6的表达水平。结果与空白对照组比较,曲古抑菌素A或香烟烟雾提取物均可显著上调巨噬细胞株RAW264.7的TNF-α、IL-6表达;应用姜黄素或地塞米松干预,可抑制炎症因子TNF-α和IL-6的表达上调;联合应用姜黄素与地塞米松具有更强的抗炎作用。结论姜黄素可抑制香烟烟雾提取物导致的巨噬细胞炎症因子TNF-α、IL-6分泌增加,可增强地塞米松的抗炎作用,其作用机制可能与姜黄素活化组蛋白去乙酰化酶有关。
Objective To investigate the anti-inflammatory effect of curcumin on RAW264. 7 macrophages activated by cigarette smoke extract. Methods RAW264. 7 macrophage cell line was cultured in medium with different concentrations of cigarette smoke extract or curcumin,and then the appropriate concentration was screened by MTT assay. Trichostatin A( a histone deacetylase inhibitor) or cigarette smoke extract stimulated RAW264. 7 cell line to simulate the inflammatory response processes of chronic obstructive pulmonary disease in vitro. Curcumin and / or dexamethasone were used to treat macrophage cell line RAW264. 7 activated by trichostatin A or cigarette smoke extract. The expression levels of inflammatory cytokines TNF-α,IL-6 in cell culture supernatants were measured by enzymelinked immunosorbent assay. Results Compared with blank control,trichostatin A or cigarette smoke extract significantly increased TNF-α and IL-6 expression of macrophage cell line RAW264. 7,while curcumin or dexamethasone inhibited this over-expression of TNF-α and IL-6. The combined application of curcumin and dexamethasone had more significant anti-inflammatory effect. Conclusion Curcumin can inhibit the increase of macrophage inflammatory cytokines TNF-α and IL-6 caused by cigarette smoke extract,and enhance the anti-inflammatory effects of dexamethasone,which may be associated with activation of the histone deacetylase.
