摘要 目的探讨缩短快速冰冻切片制片时间的新方法,进而改善冰冻切片染色效果。方法本实验对固定液进行了改进,采用甲醇和甲醛混合液作为固定剂来固定冰冻切片组织,能有效缩短快速冰冻切片的制片时间;实验中采用常规的冰冻切片方法进行切片,用改进后的固定液固定冰冻切片组织,然后常规HE染色,最后采用碱性水(30-50℃的洗衣粉水)返蓝,封片。结果使用此方法制片,切片时间可控制在3 min左右,染色时间仅用5 min,切片完整,细胞核、细胞质、细胞膜等清晰度增加;另外,此方法操作简便、经济、快速,易于掌握。结论通过对固定液的改进,采用碱性水(30-50℃的洗衣粉水)返蓝,有效地提高了快速冰冻切片的染色质量;并且制片时间缩短,方法简便快捷,能准确快速地为病理医生提供优质的冰冻染色切片。 Objective To explore a method to shorten the production time of quickly frozen sections and improve the staining quality of quickly frozen sections. Methods The fixative was improved with a mixture of methanol and formaldehyde as a fixer to fix the frozen tissue sections,which can effectively shorten the production time of frozen section. The conventional method was used for slice. The frozen sections were fixed with the improved fixative,and then stained and mounted. Results By the improved method,the slicing time was around 3 min,and the dyeing time was only 5 min. The nucleus,cytoplasm,and cell membrane were integral,and the morphology was more clear. Additionally,this method was simple,economic,fast and easy to master. Conclusion The improved method could be simple,fast,and effective to prepare the frozen sections,which quickly and accurately provide the good quality of slices for the pathologist.
