乳腺癌中肿瘤相关巨噬细胞与CXCR4表达的关系及意义 被引量:9

摘要 目的探讨乳腺癌间质中肿瘤相关巨噬细胞(tumor associated macrophages,TAMs)计数与癌细胞表达CXC家族趋化因子受体4(CXC chemokine receptor 4,CXCR4)蛋白的相关性及临床意义。方法标本均为手术切除后经常规石蜡包埋切片,采用免疫组化MaxVision法检测150例乳腺癌和50例乳腺良性病变组织中TAMs计数和CXCR4表达。结果乳腺癌组织中TAMs平均浸润密度为105.0个/HPF,中位数为87.4个/HPF。CXCR4在乳腺癌组织中表达强度明显高于乳腺良性病变组织,两组差异有统计学意义(P<0.01),且TAMs计数和CXCR4表达呈正相关(r=0.152,P=0.045)。结论 TAMs浸润和CXCR4表达与乳腺癌的发生、发展密切相关,提示TAMs浸润密度与CXCR4联合检测可作为乳腺癌临床诊断、辅助治疗的潜在靶点。 Purpose To examine the relationship between the expression of CXC chemokine receptor 4 (CXCR4) and tumor associated macrophages (TAMs) and clinical pathological significance in breast cancer. Methods MaxVision immunohistochemistry method was used for the stain of TAMs and CXCR4 in routinely paraffin-embedded sections from the samples of 150 breast carcinoma cases and 50 cases of benign breast lesions, followed by TAMs positive-counting on five botspots under high power microscope. Results The average and median invasion density of TAMs in breast cancer tissue were 105.0/HPF and 87.4/HPF. The count of TAMs and the expression of CXCR4 were significantly higher in the breast carcinoma than those in the benign breast lesion (P 〈0. 01 ) , and the count of TAMs was positively correlated with the expression of CXCR4 ( r = 0. 152, P = 0. 045 ). Conclusions The TAMs infiltration and the expression of CXCR4 may closely related to the occurrence and development of breast carcinoma, which indicating the combined detection of TAMs and CXCR4 provide a potential target for clinical diagnosis, adiuvant therapy and judgment of prognosis of breast cancer.
