子宫颈鳞状细胞癌中Klotho与Wnt10b的表达及其相关性 被引量:6

摘要 目的检测正常子宫颈上皮、子宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)以及子宫颈鳞状细胞癌(cervical squamous cell carcinoma,CSCC)组织中Klotho和Wnt10b的表达,探讨二者在子宫颈癌发生、发展过程中的作用及其相关性。方法采用免疫组化SP法检测Klotho及Wnt10b在正常子宫颈上皮、CIN及CSCC组织中的表达。结果 Klotho在正常子宫颈组织中的表达显著高于CIN及CSCC组织,其表达与子宫颈癌患者的年龄无关,与子宫颈癌FIGO分期、癌细胞分化程度以及淋巴结转移有关,差异有统计学意义(P<0.05)。Wnt10b在CSCC组织中的表达明显高于正常子宫颈上皮组织及CIN组织,其表达与分化程度、FIGO分期及淋巴结转移有关,差异有统计学意义(P<0.05)。Klotho与Wnt10b的表达呈负相关(rs=-0.416,P<0.01)。结论 Klotho和Wnt10b可能在子宫颈癌的发生、发展过程中起重要作用,联合检测二者有助于子宫颈癌发病机制的进一步研究。 Purpose To study the expression of Klotho and Wnt10b in human normal cervical epithelial tissue, cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma (CSCC), and to explore their roles in the carcinogenesis and progression of cervical cancer. Method The expression of Klotho and Wnt10b was evaluated by immunohistochemieal SP method staining in normal cervical epithelial tissue, CIN and CSCC. Result The expression of Klotho in normal cervical epithelial tissue was significantly higher than that in CIN and CSCC. Klotho expression was associated with FIGO stages, differentiation grade and lymph node metastasis ( P 〈 0.05 ) ,but unrelated to the age of patients. Positive expression of Wnt10b was remarkable higher in CIN and CSCC than that in nomal cervical epithelial tissue, which was associated with differentiation grade, FIGO stages and lymph node meatstasis ( P 〈 0.05 ). Expression of Klotho and Wnt10b were negative relationship ( rs = - 0. 416, P 〈 0. 01 ). Conclusion The expression of Klotho and Wnt10b may play a crucial role in the progress and metastasis of cervical cancer. Combinded detection of Klotho with Wnt10b provide luther study of the pathogenesis of cervical cancer.
