摘要
目的:研究三叶青黄酮对荷Lewis肺癌模型小鼠的瘤体及免疫抑制相关细胞因子表达的影响,初步探讨三叶青总黄酮逆转肿瘤免疫逃逸的可能机制。方法:匀浆接种法建立C57BL/6小鼠Lewis细胞肺癌模型,连续给药14d后观察正常组、模型对照组、三叶青黄酮高、中、低剂量组瘤体增长情况及血清中转化生长因子β1(TGF-β1)、前列腺素E2(PGE2)、环氧化酶-2(COX-2)表达含量。结果:瘤体:高剂量组、中剂量组与模型对照组相比较瘤体大小有统计学差异(P<0.05),瘤体大小依次为:模型对照组>低剂量组>中剂量组>高剂量组。血清TGF-β1表达、PGE2和COX-2含量:各实验组均明显高于正常组(P<0.01),高剂量组<低剂量组(P<0.05)。结论:三叶青黄酮高剂量组抑瘤效果较好,中剂量抑瘤效果次之,低剂量组抑瘤效果不明显。三叶青黄酮的抑瘤作用可能与降低外周血中TGF-β1的表达水平有关,可能通过影响PGE2、COX-2表达水平,从而达到逆转肿瘤逃逸,起到抗肿瘤作用。
AIM:Research on the tetrastigma hemsleyanum diels et gilg flavone(sanyeqing),which influences the tumor volume and the expression level of TGFβ1,PGE2,COX-2of LLC tumor bearing C57BL/6 mice,to explore the possible mechanism of reverse tumor immune escape function of radix tetrastigme to tumor-bearing mice.METHODS:LLC tumor bearing C57BL/6mice models were established by tumor homogenate inoculation method and were randomly divided into normal group,model control group,high dose group of the total flavonoids from radix tetrastigmae,middle dose group of the total flavones from radix tetrastigmae,low dose group of the total flavonoids from radix tetrastigmae.Stripped the tumor tissue and weighed after 14 days continuous administration.The ELISA method was used to detected the TGFβ1,PGE2and COX-2content in serum.RESULTS:Tumor volume:compared with the model control group,high-dose group,middledose group had significant difference in tumor volume(P〈0.05).Tumors size in the order:model control group〉low-dose 〉groupmiddledose group〉 high-dose group.TGF-β1,PGE2 and COX-2content in serum:after the experiment,the experimental groups were significantly higher than the normal group(P〈0.01),within groups,high-dose group〈 low-dose group(P〈 0.05).CONCLUSION:The tetrastigma hemsleyanum diels et gilg flavone highdose group had a better effect in suppressing tumor growth,the middle dose group didn't play an effective role,And the low-dose group had the most weak effect in suppressing tumor growth.The tumor suppression effect of tetrastigma hemsleyanum diels et gilg flavone may be related to the lower levels of TGF-β1expression in peripheral blood,and may decrease the level of PGE2,COX-2expression,so as to achieve reverse tumor escape,to achieve the antitumor effect.
