摘要
目的探讨大承气汤联合嗜酸乳杆菌对重型颅脑损伤(severe head injury,SHI)小鼠胃肠动力的影响。方法将75只雄性C57BL/6小鼠成功制模后按随机数字表法分为5组:假伤组、大承气汤组、嗜酸乳杆菌组、联合用药组(大承气汤+嗜酸乳杆菌)和生理盐水组,每组15只。于伤后4 h干预,干预后1、3、7 d取材,检测血浆胃动素(motilin,MTL)含量和小肠推进率,同时采用HE染色法观察脑组织形态学变化。结果 MTL含量:伤后1 d,同假伤组比较,生理盐水组明显降低(P<0.05),而嗜酸乳杆菌组明显高于生理盐水组(P<0.05);伤后3 d,联合用药组均高于生理盐水组和大承气汤组(P<0.05);伤后7 d,生理盐水组较假伤组低(P<0.05),大承气汤组明显高于生理盐水组、嗜酸乳杆菌组和联合用药组(P<0.05,P<0.01)。小肠推进率:伤后3 d,生理盐水组明显低于假伤组(P<0.01);嗜酸乳杆菌组明显高于生理盐水组和假伤组(P<0.05,P<0.01);大承气汤组和联合用药组均明显高于假伤组和生理盐水组(P<0.01)。与生理盐水组比较,大承气汤组、嗜酸乳杆菌组和联合用药组脑组织形态均有改善。结论大承气汤和嗜酸乳杆菌联合应用可通过调节MTL和肠推进功能有效改善SHI小鼠胃肠动力。
Objective To determine the effects of Dachengqi Decoction( DCQD, a set prescription of traditional Chinese herbs) combined with Lactobacillus acidophilus (LA) on the gastrointestinal motility in the mice after severe head injury (SHI). Methods A total of 75 male C57BL/6 mice were randomly divided into 5 groups (n = 15), that is, the sham trauma group, DCQD group, LA group, combination group (DCQD + LA), and saline group. The mice of the latter 4 group were inflicted to SHI and those of the first group received sham injury. In 4 h after SHI, the corresponding treatment was given to above groups. The intestinal propulsion rates were tested in 3 d after intervention, and the blood samples and brain tissues were collected in 1, 3, and 7 d after intervention. The serum plasma level of motilin (MTL) was detected by ELISA, and the brain morphology was observed by HE staining. Results In 1 d after SHI, the MTL level was significantly lower in saline group than the sham trauma group (P 〈 0. 05 ), and higher in the LA group than in saline group ( P 〈 0. 05). In 3 d after SHI, the level was significantly higher in the combination group than saline group and DCQD group ( P 〈 0. 05 ). The level was obviously lower in 7 d after SHI in saline group than the sham trauma group ( P 〈 0. 05 ), and was higher in DCQD group than saline group (P 〈 0.01 ), LA group (P 〈 0. 05 ) and combination group (P 〈0. 01 ). After 3 d post-trauma, the small intestinal propulsion rates was significantly lower in the saline group than in the other 4 groups ( P 〈 0. 01 ), and that of the combination group was significantly higher than the sham trauma group (P 〈 0. 01 ). Compared with the saline group, the brain tissue morphology was obviously improved in the DCQD group, LA group and combination group. Conclusion DCQD combined with LA effectively improves gastrointestinal dynamics in SHI mice by adjusting MTL and intestinal propulsion function.
