摘要
目的:探讨兔VX2肺癌模型多层螺旋CT灌注扫描图像与肿瘤乏氧标志表达和肿瘤乏氧区间的相对关系。方法:17只新西兰大白兔,肺部原位成瘤后行动态CT灌注扫描,应用Perfusion 3灌注软件行灌注图像重建,在肿瘤高灌注区、低灌注区各选取3个感兴趣区做比较,肿瘤组织标本行免疫组化,检测缺氧诱导因子-1α(hypoxia-inducible factor-1alpha,HIF-1α)、血管内皮生长因子(vascular endothelial growth factor,VEGF)和肿瘤微血管密度(microvessel density,MVD)的表达。应用SAS 9.0统计软件包进行数据统计分析。结果:高灌注区与低灌注区比较,HIF-1α的表达差异有统计学意义,P<0.01;血容量差异有统计学意义,P<0.01。在高灌注区,HIF-1α和血容量有明显相关性,r=-0.75,P<0.01;HIF-1α和血流量亦有明显相关性,r=-0.50,P<0.05。在低灌注区,HIF-1α与血容量(r=-0.50,P<0.05)、血流量(r=-0.49,P<0.05)和表面通透性(r=-0.53,P<0.05)均有相关性。结论:初步提示,CT灌注图像高、低灌注区和乏氧区有一定的相关性。
OBJECTIVE:To analyze the parameters of MSCT perfusion scan images and the expression level of hypoxia markers,explore the interval relative relationship of MSCT perfusion scan images and tumor hypoxia areas.METHODS:Seventeen female New Zealand white rabbits were implanted with the VX2tumor cell in lung.MSCT plain scan and perfusion scan were performed.With the help of perfusion 3software,perfusion scan images were rebuilded.In the high and low perfusion image,we selected three region of interest(ROI)respectively.After CT perfusion,the rabbit's tumor specimen was harvested.H E staining and hypoxia-inducible factor-1alpha(HIF-1α),vascular endothelial growth factor(VEGF),microvessel density(MVD)immunohistochemical staining were carried out.SAS 9.0statistical package was used for analysis.RESULTS:To compare high perfusion region and low perfusion region,using the blood flow three-colorimage(red,green,blue)as the discriminating standard,HIF-1α,VEGF had obvious difference in statistic between high and low perfusion(all P〈0.05).HIF-1αwith BV,BF all had negative correlation in statistics(r=-0.75,P〈0.01;r=-0.50,P〈0.05)in high perfusion region,difference from HIF-1αwith BV,BF,PS all had negative correlation in statistics in low perfusion region(r were-0.50,-0.49and-0.53,all P〈0.05).CONCLUSION:The results preliminary prompts that CT perfusion images and hypoxic areas have some relevance in high and low perfusion region.
