海尔福对铅中毒小鼠模型保健效果的研究 被引量:3

Study on health care effect of Haierfu syrup on lead poisoning mouse model

摘要 目的:探讨中药海尔福口服液对铅中毒小鼠模型的排铅保健效果,为研究铅中毒儿童排铅保健作用机理及应用提供实验依据。方法:用乙酸铅水溶液行小鼠腹腔注射染毒,将染毒模型组分成不治疗组(即模型组)和治疗组,同时按大、小计量、又分成治疗1组和治疗2组。将正常小鼠设为对照组。治疗组用大小不同剂量口服液进行灌胃测定小鼠血红蛋白(Hb)。实验结束后,从眼球取血,分离血清,测定丙氨酸氨基转移酶(ALT)、肌酐、总蛋白含量,取肝、肾、睾丸送病理检查,取大脑做成10%脑匀桨,测定O2·清除率和脑铅含量。结果:对照组、模型组、治疗1组、治疗2组测定结果依次排列,造模前Hb各组比较差异无统计学意义(P>0.05);造模后(治疗前)Hb与对照组比较,差异有统计学意义(P<0.01);治疗后Hb对照组高于其他3组,差异有统计学意义(P<0.01),同时模型组低于治疗组(P<0.05或P<0.01);ALT,对照组低于其他3组,差异有统计学意义(P<0.01);肌酐,模型组高于其他3组(P<0.01);血清总蛋白,模型组低于对照组,差异有统计学意义(P<0.05);脑O2·清除率,与对照组比较,对照组清除率明显升高,差异有统计学意义(P<0.05或P<0.01);脑铅与模型组比较,差异有统计学意义(P<0.05或P<0.01);病理形态学研究表明,肝、肾、睾丸中毒组均有一定程度的损害,治疗后有明显好转。结论:实验表明,铅染毒小鼠后,使造血、肝、肾、睾丸组织细胞形态和功能及脑抗氧化功能等受到明显损害,治疗后通过排铅,使脑铅下降,损害有明显减轻,起到保健作用。 Objective: To explore the removing lead health care effect of Haierfu syrup on lead poisoning mouse model,provide experimental basis for researching the mechanism and application. Methods: Lead acetate solution was injected into the mice transperitoneally,then the mice were divided into model group( without treatment) and treatment group,the mice in treatment group were divided into treatment 1 subgroup and treatment 2 subgroup according to the dose of Haierfu syrup; normal mice were selected as control group; the mice in treatment group were treated with intragastric administration of Haierfu syrup( different doses),then the level of hemoglobin was detected;after the experiment,blood samples were obtained from eyeballs and the serum was isolated; the contents of alanine aminotransferase( ALT),creatinine and total protein were detected; pathological examination on liver,kidney and testis was performed; cerebral tissue was obtained for preparing 10% brain homogenate,the clearance rate of O2·and cerebral lead content were measured. Results: Before establishing model,there was no statistically significant difference in hemoglobin level among control group,model group,treatment 1 group and treatment 2 group( P〈0. 05); after establishing model( before treatment),compared with control group,there was statistically significant difference( P〈0. 01); hemoglobin level in control group after treatment was statistically significantly higher than those in the other three groups( P〈0. 01),hemoglobin level after treatment in model group was statistically significantly lower than that in control group( P〈0. 05); ALT level in control group was statistically significantly lower than those in the other three groups( P〈0. 01); creatinine level in model group was statistically significantly higher than the other three groups( P〈0. 01); serum total protein level in model group was statistically significantly lower than that in control group( P〈0. 05 or P〈0. 01);; the clearance rate of cerebral O2·in control group increased significantly( P〈0. 05 or P〈0. 01); compared with model group,there was statistically significant difference in cerebral lead content( P〈0. 05 or P〈0. 01); pathomorphological study showed that there were a certain injuries in liver,kidney and testis in lead poisoning group,which were improved significantly after treatment. Conclusion: The experiment showed that lead could damage cell morphologies and functions of hematopoiesis,liver,kidney,testis and cerebral antioxidant function obviously,after treatment of removing lead,the level of cerebral lead decreased,the injuries were relieved.
