一株中性内切纤维素酶产生菌的分离及鉴定 被引量:1

摘要 利用刚果红染色法从偏碱性土壤中分离产内切纤维素酶的菌株,通过内切纤维素酶(CMC酶)的发酵液酶活和酶促反应最适pH,复筛得到1株中性内切纤维素酶产生菌A8。经形态学观察、生理生化特征分析及分子生物学鉴定,A8菌株属于蜡状芽孢杆菌(Bacillus cereus)。其在50℃、pH7.0发酵96h条件下发酵液的中性内切纤维素酶酶活为141.70U/mL,是一株在食品加工中具有开发潜力的产中性内切纤维素酶菌株。 The bacterial strains producing endo-cellulase were isolated from alkalescent soil by using the method of Congo red dye.In these strains, a bacterial strain A8 with high neutral endo-cellulase activity was obtained by analyzing of the endo-cellulase activity(CMCase activity)and the optimal enzymatic reaction pH.According to morphology,biochemical and physiological characterization, and 16S rDNA sequence analysis, A8 was identified as Bacillus cereus, After incubation for 96h,the neutral endo-cellulase of shaking culture of A8 was 141.70U/mL with the temperature of 50℃ and the pH of 7.0.A8 was a neutral endo-cellulase producing strain with development potential in the food manufacture industry.
