miR-15a对肝癌HepG2细胞增殖和DLK1表达的影响 被引量:3

摘要 目的探讨miR-15a对肝癌中DLK1表达的调节作用和对肝癌HepG2细胞增殖的影响。方法常规培养肝癌细胞系HepG2,经脂质体转染miR-15a模拟物mimic和抑制物inhibitors及各自阴性对照片段NC48 h后,用RT-PCR和Western blot检测DLK1基因及蛋白的表达水平,MTT和流式细胞术(FCM)检测细胞增殖和周期情况。结果 RT-PCR和Western blot结果显示转染miR-15a mimic后DLK1的mRNA和蛋白表达量降低(P<0.05),而转染miR-15a inhibitors后DLK1的mRNA和蛋白表达量升高(P<0.05);MTT结果显示转染miR-15a mimic后细胞的增殖能力明显减慢(P<0.05),而转染miR-15a inhibitors后细胞增殖能力加快(P<0.05)。FCM结果显示转染miR-15a mimic后细胞抑制在G1期。结论过表达miR-15a能抑制DLK1的mRNA和蛋白表达,并抑制肝癌HepG2细胞的增殖。
