布美他尼后处理对大鼠创伤性颅脑损伤后NKCC1表达及脑含水量的影响 被引量:3

摘要 目的探讨布美他尼后处理对大鼠创伤性颅脑损伤后Na+-K+-2Cl-协同转运蛋白1(NKCC1)表达及脑含水量的影响。方法雄性SD成年大鼠90只,体重220-300 g。采用随机数字表法,将大鼠分为3组:假手术组(S组)、脑创伤组(TBI组)和布美他尼后处理组(TBI-B组)。采用自由落体撞击法制备创伤性颅脑损伤模型。S组接受同样的操作但不接受撞击,TBI-B组于创伤后30 min给予布美他尼30 mg/kg,TBI组给予等量的生理盐水。TBI组和TBI-B组在创伤后12 h测动脉压并取动脉血行血气分析,于创伤后2、12、24 h处死大鼠并取脑,测定皮质NKCC1的表达和脑含水量。结果与S组比较,TBI组和TBI-B组NKCC1表达水平和脑含水量在创伤后12、24 h升高(P〈0.05,P〈0.01);与TBI组比较,TBI-B组NKCC1表达和脑含水量在创伤后12 h、24 h降低(P〈0.05,P〈0.01)。结论布美他尼后处理可减轻成年大鼠创伤性脑损伤,其机制可能与布美他尼减少大鼠脑组织NKCC1表达有关。
