摘要
目的探讨泽兰水煎剂对实验性小鼠溃疡性结肠炎急性期结肠黏膜病理的影响。方法健康雄性昆明小鼠60只,按随机数字表法分为对照组、模型组和泽兰组,每组20只。对照组自由饮用蒸馏水+生理盐水灌胃,模型组饮用含5%葡聚硫酸钠(DSS)的蒸馏水+生理盐水灌胃,泽兰组饮用含5%DSS的蒸馏水+泽兰水煎剂灌胃。实验结束时处死小鼠,取近肛门2 cm处结肠组织,制作病理组织切片,光镜下观察。结果模型组、泽兰组均成功建立小鼠溃疡性结肠炎急性期模型。实验结束时对照组、泽兰组小鼠体重均明显大于模型组(P<0.05),但对照组小鼠体重与泽兰组比较,差异无统计学意义(P>0.05)。实验结束时对照组、泽兰组小鼠体重均明显高于实验前(P<0.05),实验结束时模型组小鼠体重明显低于实验前(P<0.05)。对照组和泽兰组小鼠结肠标本大体形态和组织形态学评分均明显低于模型组(P<0.05);而泽兰组结肠标本大体形态和组织形态学评分与对照组比较,差异均无统计学意义(P>0.05)。结论泽兰能明显减轻DSS所致小鼠溃疡性结肠炎的急性期病理改变。
Objective To investigate the effect of lycopus decoction on the pathological changes of colonic mucous membrane in mice with ulcerative colitis( UC) in acute phase. Methods Sixty healthy male Kunming mice were divided into control group,model group and lycopus group according to the random number table,with 20 mice in each group. The mice in the control group freely drank distilled water and were gavaged with normal saline; In the model group,instead of 5% dextran sulfate sodium( DSS) solution plus normal saline by gavage; In the lycopus group,replaced by 5% DSS solution plus lycopus decoction by gavage. The animals were killed at the end of experiment,and then the colon tissue sampling which was 2 cm near anus was performed,and then the pathological sections were observed with light microscope. Results The mouse model of ulcerative colitis in acute phase was established successfully both in the model group and lycopus group. At the end of experiment,the body weight of mice in the control group or lycopus group was heavier than that in the model group( P〈0. 05),but there was no significant difference in the body weight of mice between control group and lycopus group( P〉0. 05). The body weights of mice in the control group and lycopus group at the end of experiment were heavier than those before experiment( P〈0. 05),the body weight of mice in the model group at the end of experiment was lighter than that before experiment( P〈0. 05). The scores of gross morphology and histomorphology of colonic samples in the control group and lycopus group were significantly lower than those in the model group( P〈0. 05). There were no significant differences in the scores of gross morphology and histomorphology of colonic samples between lycopus group and control group( P〉0. 05). Conclusion Lycopus can significantly alleviate the acute pathological changes of colonic mucosa membrane in mice with DSS-induced UC.
