摘要
运用传统育种方法 ,用整合有人凝血因子 ( h F )的转基因小鼠进行近交和远交。通过选育 ,在其后代中出现 4只双重杂合子 ( 3♀ ,1 )及 2只纯合子小鼠 ( 2♀ )。收集雌鼠乳汁 ,用 ELISA测定其中 h F 蛋白含量。结果表明 ,相对于单纯杂合子小鼠 ,纯合子小鼠的表达量无增高 ( P>0 .0 5) ,而双重杂合子小鼠的表达量有显著提高 ( P<0 .0 1 )。结果提示 ,选育双重杂合子转基因后代小鼠 ,是一条提高转基因动物外源基因表达水平的有效途径。
Transgenic mice with human factor Ⅸ(hFⅨ) gene were bred.Through hybridization,four double heterozygous(3 ♀,1 ) and two homozygous(2 ♀) transgenic mice were selected among offspring.The milk was collected during lactating and the hFⅨ protein was measured by an ELISA method.The data showed that the hFⅨ protein expression was not significantly different between homozygotes and heterozygotes(P>0 05),and their expression in double heterozygous mice was significantly higher than that in heterozygotes(P<0 01).These results indicated that the selection of double heterozygotes among offspring may be a useful way to improve the expression of exogenous genes in the transgenic animals.
