摘要
对柿品种'禅寺丸'未减数花粉(2n花粉)的鉴定、筛选及其生活力恢复进行了研究,结果表明:(1)经0.9 mol/L甘露醇CPW溶液吸胀2h后,正常花粉直径为50 μm左右,而未减数花粉为65 μm左右,未减数花粉直径是正常花粉的1.3倍。(2)未减数花粉比率约为3.8%,通过61μm的不锈钢细胞筛筛选,纯度可达84%。(3)禅寺丸花粉经吸胀处理后,花粉发芽率有所下降,利用20%的蔗糖溶液洗涤可基本恢复其生活力。
Studies were carried out on the identification,screening and recovery of viability of unreduced pollen (2n pollen) in Japanese 'Zenjimaru' persimmon variety Diospyros kaki L. f .The results showed that 1 The diameter of normal pollen and unreduced pollen was 50 μm and 65 μm,respectivelyafter imbibed about 2 hours in CPW solution added 0.9 mol/L mannitol. Unreduced pollen was 1.3 times to normal one in diameter.2Unreduced pollen percentage was 3.8% naturally and could be increased to 84% after screened by stainless steel mesh.3The viability of pollen declined after imbibed but could recover partly by 20% sucrose solution.
