巴中栀子遗传多样性的ISSR分析 被引量:8

ISSR analysis on the genetic diversity of Fructus Gardeniae from Bazhong producing area

摘要 目的:运用ISSR方法分析巴中、江津、纳溪3个不同产地栀子的亲缘关系及巴中产栀子种质资源的遗传多样性。方法:根据优化的ISSR-PCR反应体系,筛选出12个引物对3个不同产地栀子的DNA进行扩增,并应用NTSYS2.10e软件,对所得数据进行分析处理。得到3个不同产地栀子的遗传相似系数和遗传距离,并绘制出样品相似性的UPGMA聚类图和主坐标分析的二维、三维散点图。结果:巴中30年生栀子群,巴中3年生栀子群,江津栀子群,纳溪栀子群,巴中庭院栽培群亲缘关系较近;但各群体间又存在明显遗传变异。结论:巴中、江津和纳溪产栀子的遗传多样性较明显,种质间存在一定的遗传分化;从基因层面上解释了巴中栀子在药材外观、大小、色泽和有效成分的含量、特征指纹图谱、药效评价指标等诸多方面品质优异的可能性。 OBJECTIVE By using ISSR to research Fructus Gardeniae kinship of three different areas in Bazhong, Jiangjin, Naxi, and explore germplasm genetic diversity of Fructus Gardeniae in Bazhong. METHODS According to the optimization of ISSR-PCR reaction system, 12 primers were screened to amplify three different origin Gardenia DNA, software NTSYS 2. 10e was applied to analyze the data. The systematic diagram of three different origin Gardenias and its genetic distances were ob- tained, the sample similarity UPGMA cluster map and principal coordinate analysis of two-dimensional, three-dimensional scat- ter plot were drawn. RESULTS Bazhong 30 years old gardenia group, and Bazhong 3 years old gardenia group, jiangJin garde- nia group, Naxi gardenia group and Bazhong garden cultivation had closer relationship. But there were obvious genetic variation among populations. CONCLUSION There are obvious genetic diversity among gardenia in Bazhong, Jiangjin and Naxi produ- cing area. And now the Bazhong Gardenia herbs' appearance, size, color and content of active ingredient, characteristic finger- print, efficacy evaluation, and many other aspects of superior quality possibility and reality can be explained from the genetic level.
