摘要
目的:研究粗壮唐松草提取物的体外抗肿瘤活性。方法:采用MTT法测定粗壮唐松草提取物各部位对NCI-H460细胞、HepG-2细胞、MCF-7细胞、Hela细胞和S180细胞的抑制作用,以筛选抗肿瘤活性部位;采用MTT法考察生物碱部位对HepG-2细胞和Hela细胞抑制作用的特性;采用流式细胞术检测生物碱部位诱导HepG-2细胞和Hela细胞凋亡的作用。结果:粗壮唐松草生物碱部位体外对5种肿瘤细胞的IC50分别为1.05 g·L-1(NCI-H460)、0.127 g·L-1(HepG-2)、0.976 g·L-1(MCF-7)、0.320 g·L-1(Hela)和0.404 g·L-1(S180),抑制作用均优于其他各部位,且对HepG-2细胞和Hela细胞抑制作用呈现良好的时间与浓度依赖性;粗壮唐松草生物碱部位能诱导HepG-2细胞和Hela细胞的凋亡。结论:粗壮唐松草生物碱部位能显著抑制HepG-2和Hela等肿瘤细胞的生长,具有诱导凋亡的作用,为粗壮唐松草提取物的抗肿瘤活性部位。
OBJECTIVE To investigate the anti-tumor effect of extracts of Thalictrum robustum Maxim. in vitro. METH- ODS The cytotoxicity of NCI-H460 ceils, HepG-2 cells, MCF-7 cells, Hela cells and S180 cells were tested by cell inhibitory assay to screen out the effective extract of Thalictrurn robustum Maxim.. The inhibitory effects of the alkaloids extract on HepG-2 and Hela ceils were tested by MTT. FCM assays were used to detect cell apoptosis induced by the alkaloids extract. RESULTS The alkaloids extract of Thalictrum robusturn Maxim. inhibited the proliferation of these five kinds of tumor cells, and ICs0 values for them were 1.05 g·L -1 (NCI-H460), 0. 127 g·L -1 (HepG-2), 0. 976 g·L -1 (MCF-7), 0. 320 g·L -1 (Hela) and 0. 404 g·L -1 (S180), respectively. The inhibitory effect of the alkaloids extract showed a time- and dose-dependent man- ner. The tumor cell apoptosis induced by the alkaloids extract was detected. CONCLUSION The anti-tumor effective extract of Thalictrum robusturn Maxim. in vitro was the alkaloids extract, which showed significant anti-tumor activities in vitro, and in- duced tumor cell apoptosis.
