以2-氨基-4′-氟二苯甲酮为起始原料,经成喹啉环、还原、溴代、制成膦叶立德和Wittig-Hornor反应、脱保护、缩合,最后水解成盐得到降血脂药物匹伐他汀钙,总收率为24.8%。产品结构经IR和1H NMR确认。
Pitavastatin calcium is an important lipid regulator which is usually prepared by resolution of racemic or chiral synthesis. The authors have developed an improved process for the synthesis of pitavastatin calcium starting from 2-amino-4'-fluorobenzophenone in several steps in overall yield of 24.8%. The structure of the title compound was confirmed with IR and 1H NMR spectra.
