摘要
利用农杆菌介导玉米愈伤组织遗传转化,研究了不同影响因子对玉米遗传转化效率的影响。结果表明,相比自交系H99,玉米杂交组合H99×A188更适宜作为玉米遗传转化的受体材料;继代6d后的胚性愈伤组织最适宜农杆菌转化;农杆菌菌株EHA105的侵染效果好于EHA101;抗性愈伤组织筛选培养基中除草剂筛选的临界质量浓度为4.0mg/L;头孢霉素质量浓度在500mg/L时适合进行除菌处理,且对植株再生影响较小。
The embryogenic callus of maize transformation was generated by Agrobacterium tumefaciens.The result showed that the resistant callus regenerated was influenced by the genotype,time of callus subculture,Agrobacterium strains,herbicide concentration and antibacterial agent concentration.The genotype was the main factor affecting the genetic transformation of maize,and the calli subcultured for six days were optimum for agrobacterium infection.The Agrobacterium tumefacien strain EHA105 was better than EHA101 for infection.In selection medium,the highest concentration of herbcide was 4.0 mg/L and the suitable concentration of antibiotics was 500 mg/L,which had the lowest influence on the plant regeneration.
