卡因酸对大鼠学习记忆及胆碱酯酶和S^(100)β表达的影响 被引量:1

摘要 目的 探讨卡因酸对大鼠学习记忆能力的影响 ,观察其基底前脑皮质及海马乙酰胆碱酯酶和S10 0 β表达的变化。方法 大鼠腹腔注射卡因酸为实验组 ,注射磷酸缓冲生理盐水为对照组。用Y迷宫测试大鼠学习记忆 ,乙酰胆碱酯酶 (acetyl cholineesterase ,AchE)化学染色法测定AchE表达水平的变化 ,免疫组化法检测S10 0 β表达水平的变化。 结果 实验组大鼠的学习记忆能力明显低于对照组 (P <0 .0 5 ) ,AchE表达水平明显低于对照组 (P <0 .0 1) ,S10 0 β表达水平明显高于对照组 (P<0 .0 1)。结论 腹腔注射卡因酸可以损害大鼠的学习记忆能力 ,减少AchE的表达 ;过高浓度的S10 0 Objective To study effect of kainite on the learning and memory abilities of rats and to observe the change of AchE and S 100 β in hippocampi of rats. Method The rats were divided into two groups: the experimental group was made with the intraperitoned administration of kainite, the control group was made with phosphate buffer solution (PBS) in some way. All rats in two groups were measured with Y mize tests and AchE was detected by histochemistry of AChE, S 100 β was observed by immunohistochemical assay. [WT5”HZ]Results The experiment showed that the learning and memory abilities of experimental rat were worse than that of control rat (P<0.05).The AchE contents of in the hippocampi and basal forebrain region of experimental rat was significantly decreased than that of control rat (P<0.01).The S 100 β contents of in the hippocampi of experimental rat was significantly increased than that of control rat (P<0.01). Conclusion The learning and memory abilities of rats could be impaired by the intraperitoned administration of kainite. Elevated S 100 β content might be involved in the pathophysiologic course of the learning and memory disorders by kainite.
