摘要
目的:探讨胡椒碱对戊四氮致痫大鼠海马氨基酸递质的影响。方法:48只成年SD大鼠随机分为对照组、模型组和实验组,模型组和实验组大鼠腹腔注射戊四氮60mg/kg,诱导癫痫发作。实验组于注射戊四氮之前1h经腹腔注射胡椒碱60mg,并观察1h。然后处死大鼠取脑,应用柱前衍生化高效液相色谱荧光法测定戊四氮致癫痫大鼠脑海马谷氨酸和γ氨基丁酸的含量。结果:实验组海马谷氨酸和GABA含量与模型组相比差异均有显著性(P<0.01)。结论:胡椒碱可以通过抑制谷氨酸的释放,促进GABA的释放,在癫痫发作中发挥保护作用。
Objective:To explore the effect of piperine on glutamate andγ-Aminobutyric acid of hippocampus in seizures rats induced by pentylenetetrazole(PTZ).Methods:Forty-eight adult SD rats were randomly divided into the control group(CG),the model group(MG)and the test group(TG).A dose of PTZ(60mg/kg)was intraperitoneally injected into the rats of the model group andthe test group and seizures were induced.The rats of the test group were administered piperine(60mg)intraperitoneally 1hbefore injecting of PTZ.The influence of glutamate andγ-Aminobutyric acid of hippocampus was studied by HPLC-fluo rescence with the pre-column derivation method.Results:The contents of glutamate andγ-Aminobutyric acid of hippocampus in the test group were obviously different from those of model groups(P<0.01).Conclusion:Piperine can exert the protective effect on seizure and restrain the release of both glutamate andγ-aminobutyric acid.
