慢病毒介导的ATF5基因沉默及其对荷人卵巢癌裸鼠移植瘤增殖的影响

摘要 目的观察慢病毒介导的人转录激活因子5(ATF5)基因沉默效果,及其对荷人卵巢癌裸鼠移植瘤增殖的影响。方法建立荷人卵巢癌裸鼠模型30只,随机分为实验组、阴性对照组、空白对照组各10只,分别瘤内注射ATF5-siRNA慢病毒、空载慢病毒、PBS。观察移植瘤生长情况,绘制移植瘤生长曲线。4周后处死裸鼠,取出移植瘤组织并称重,计算实验组抑瘤率。采用RT-PCR和Western blot方法检测各组ATF5 mRNA和蛋白相对水平,TUNEL法检测各组移植瘤组织凋亡指数(AI)。结果实验组移植瘤生长速度明显小于阴性对照组和空白对照组,其抑瘤率为49.60%。实验组ATF5 mRNA及蛋白相对表达量、AI分别为0.40±0.09、0.46±0.05、34.63%±1.91%,阴性对照组分别为0.67±0.07、0.72±0.03、6.04%±0.19%,空白对照组分别为0.80±0.07、0.82±0.05、5.07%±0.20%;实验组与阴性对照组、空白对照组比较,P均<0.01;空白对照组与阴性对照组比较,P>0.05。结论慢病毒介导的ATF5-siRNA能抑制荷人卵巢癌裸鼠移植瘤中ATF5基因的表达,并明显抑制移植瘤的生长、促进其凋亡。
