摘要
目的探讨阿霉素对大肠癌SW480细胞周期及周期相关蛋白表达的影响。方法体外培养SW480细胞,随机分为A(空白对照组)、B、C、D、E组,阿霉素浓度分别为0、2.5、5、10、20μg/mL。采用MTT法测定各组细胞培养12、24、48、72 h后的细胞生长抑制率;流式细胞术分析各组细胞培养48 h后细胞周期;Western blot法测定各组细胞培养48 h后SW480细胞周期相关蛋白Cyclin B1、Cyclin A表达水平。结果阿霉素抑制SW480细胞增殖具有浓度和时间依赖性。不同浓度阿霉素作用于SW480细胞48 h后,D、E组G0/G1期细胞百分率低于A、B、C组,SubG1期细胞百分率、凋亡指数(AI)高于A、B、C组,P均<0.05;与A组比较,B、C组Cyclin B1表达量增高(P均<0.05),D、E组表达量下降(P均<0.05);随着阿霉素浓度增加,Cyclin A的表达量逐渐降低(P<0.05)。结论阿霉素可将SW480细胞周期阻滞于G0/G1期,影响细胞周期及Cyclin B1、Cyclin A的表达。
Objective To investigate the effect of adriamycin on the cell cycle and cycle-related protein expression of colorectal cancer SW480 cells.Methods SW80 cells cultured in vitro were randomly divided into groups A (blank control group), B, C, D and E, which were treated with different concentrations of adriamycin (0μg/mL, 2.5μg/mL, 5.0μg/mL, 10 μg/mL and 20 μg/mL).After being cultured for 12, 24, 48 and 72 h, the growth inhibition rate of each group was detected by using methyl thiazolyl tetrazolium (MTT) method.After being cultured for 48 h, the cell cycle of SW480 cells in each group was analyzed by flow cytometry , and the expression of cycle-related proteins (cyclin B1, cyclin A) of each group was detected by Western blotting .Results The inhibitory rate of SW480 cell growth affected by adriamycin was in a dose-and concentration-dependant manner .After being treated by different concentrations of adriamycin for 48 h, the percentage of cells in the G 0/G1 phase of the groups D and E was lower , the percentage and apoptosis index in the SubG1 phrase of groups D and E were higher than those of groups A , B and C (all P〈0.05).The expression level of cy-clinBl in the group B and group C was higher than that of group A , but the expression level of cyclinBl in the group D and group E was lower than that of group A (all P〈0.05).The expression of CyclinA gradually decreased with the increasing concentration of adriamycin (P〈0.05).Conclusion Adriamycin can block the cell cycle at the G 0/G1 phase of SW480 cells, and effect the expression of cyclin B 1 and cyclin A .
