摘要
目的:采用高效液相色谱法测定牛樟芝中腺苷的含量。方法色谱柱:Agilent Zorbax SB-C18柱(4.6 mm ×250 mm,5μm),流动相:乙腈-0.01 mol/L磷酸二氢钾溶液(6∶94),流速:1.0 ml/min,检测波长:260 nm,柱温:30℃,进样量:10μl。结果腺苷在2.696~67.40 g/ml范围内与其峰面积呈现良好的线性关系(r=0.9999),平均回收率为99.89%,相对标准偏差为1.71%(n=6)。结论该方法简便准确,灵敏度高,重复性好,为控制牛樟芝的质量提供了科学依据。
Objective To detect the Adenosine content in Antrodia Camphorata using high performance liquid chromatography (HPLC). Methods The chromatographic column was Agilent Zorbax SB-C18 column (4. 6 mm × 250 mm, 5 μm); the mobile phase was acetonitrile-0. 01 mol·L-1 KH2PO4 solution (6∶ 94); the flow rate was 1. 0 ml/min-1; the detection wavelength was 260 nm;the column temperature was set at 30℃;the injection volume was 10 μl. Results A good linear relationship between the peak area and Adenosine concentration was observed within the range of 2. 696-67. 40 mg/ml (r=0. 9999). The average recovery was 99. 89% with 1. 71% of relative standard devia-tion (n=6). Conclusion This method is accurate and has good sensitivity and reproducibility, and can provide scien-tific basis for controlling the quality of Antrodia camphorata.
