Dicer在RNA沉默中的作用

摘要 RNA沉默是一种在真核生物中广泛存在的防御机制,它的一个主要作用是抗病毒。为了阐明这个机制,很多植物病毒的功能蛋白都被用来做RNA沉默抑制子的研究。RNA III聚合酶家族的Dicer在基因沉默机制中发挥关键的作用,不同的物种含有不同数量和种类的Dicer,可以将外源的mRNA切割成21-26nt的小片段,从而阻断外源基因的翻译途径,达到基因沉默的目的。病原物在进化的过程中,编码沉默抑制子蛋白,对抗寄主的基因沉默。沉默抑制子通过与寄主的蛋白或外源RNA等互作,抑制寄主的基因沉默。关于Dicer与沉默抑制子的研究将给更好地解析基因沉默机制提供理论基础。
