摘要
目的观察孕鼠羊水栓塞模型实验前后血清补体C3、C4和B因子的动态变化,探究羊水栓塞引发的类过敏反应。方法取健康妊娠晚期SD大鼠40只,随机分为两组,每组20只。两组孕鼠麻醉后从左侧股静脉取血,再暴露子宫抽取羊水。按2.5ml/kg由右侧股静脉缓慢注入自体羊水作为实验组,注入等剂量生理盐水作为对照组。注射1h后从右心房取血,将所取血样离心后分离血清,用ELISA法检测血清中补体C3、C4和B因子含量。结果羊水注射1h后血清补体C3、C4和B因子降低,差异有统计学意义(P<0.05)。结论孕鼠羊水栓塞补体系统被过度激活,消耗了补体成分,引发类过敏反应。
Objective The aim of the study was to investigate the dynamic change and significance of serum complement 3, complement 4 and B factor in amniotic fluid embolism in pregnant rats. Methods 40 healthy female SD rats in late pregnancy were randomly assigned into two groups. Venous blood was abstracted from the left femoral vein and amniotic fluid was extracted from the exposed uterus after anesthesia. Amniotic fluid was transfused through the right femoral vein to make the model of amniotic fluid embolism and saline was transfused as the control. Venous blood was abstracted from the right atrium one hour later. Serum was separated from venous blood by centrifuge and complement 3, complement 4 and B factor were measured by ELISA. Results Serum complement 3, complement 4 and B factor decreased after amniotic fluid embolism and the difference was statistically significant (P〈0.05). Conclusion The complement system is excessive activation and the complement components are consumed after amniotic fluid embolism that is anaphylactoid reaction.
