miR-143、miR-145及血管紧张素II在人冠状动脉粥样硬化斑块中的表达 被引量:4

The expression of miR-143,miR-145 and angiotensin II in human atherosclerotic coronary artery

摘要 目的探究miR-143、miR-145及血管紧张素II(AngII)在人冠状动脉粥样硬化斑块中的表达情况。方法采用12例冠状动脉粥样硬化解剖病例的硬化冠状动脉及斑块和13例非心源性死因病例的冠状动脉血管的石蜡包埋组织,提取组织RNA,逆转录后运用qPCR技术检测miR-143、miR-145在病变冠脉组织和正常冠脉组织中的表达情况,同时运用免疫组织化学染色技术半定量AngII在病变冠脉组织和正常冠脉组织中的表达情况。结果冠状动脉粥样硬化组中miR-143、miR-145的表达量分别为(1.16±0.36)、(3.22±1.30),较对照组的(5.21±2.86)、(13.09±6.06)下降,差异均有统计学意义(P<0.01)。AngII主要表达于粥样硬化冠状动脉的巨噬细胞及成纤维细胞中,其表达量在冠状动脉粥样硬化组中为(7.09±0.71),较对照组的(42.75±29.59)下降,差异有统计学意义(P<0.01)。对照组不同死因病例间miR-143、miR-145和AngII表达差异无统计学意义。结论 miR-143、miR-145及AngII在人冠状动脉粥样硬化斑块中表达下调。 Objective We aimed at investigating the expression of miR-143, miR-145, and their downstream protein catalysate angiotensin Ⅱ(AngⅡ) in human atherosclerotic coronary artery. Methods The expression of miR-143 and miR- 145 in formalin fixed paraffin-embedded coronary arteries from twenty-five individuals (n=25) was determined by qRT- PCR. Twelve patients died from acute coronary syndrome were classified as AP group (n=12), while thirteen patients died from other causes were classified as non-AP group (n=13). The concentration of AngII in atherosclerotic arteries was semi-quantified by immunohistochemistry. Results Our data showed that, compared with normal arteries, miR-143 and miR-145 were downregulated in atherosclerotic coronary arteries [ (1.16±0.36) vs (5.21±2.86),P〈0.01 ; (3.22±1.30) vs (13.09±6.06),P〈0.01 ].Besides, AngⅡ positive signals, mainly located in macrophages and fibroblasts in atherosclerotic arteries,were significantly lower than those in normal arteries I (7.09±0.71) vs (42.75±29.59) ; P〈0.01 ]. In control group, there was no significant difference of the expression of miR-143,miR-145 and AngⅡ among different causes of death. Conclusion miR-143, miR-145 and AngⅡ were downregulated in human atherosclerotic plaques.
