5种快速提取黑曲霉基因组DNA方法的比较 被引量:2

摘要 黑曲霉是发酵工业广泛应用的重要菌种,可产生葡萄糖氧化酶、纤维素酶、淀粉酶、果胶酶、葡萄糖酸、柠檬酸等重要物质,且产量较高。通过基因工程方法进行表达已成为提高产物产量主要的方法之一,而从黑曲霉中提取高纯度的DNA是基因克隆的前提条件。黑曲霉属于丝状真菌,因其细胞壁多糖与几丁质含量较高,细胞壁结构坚固,提取DNA的过程中破壁困难,对提取DNA的纯度与质量会造成一定的影响。目前,提取黑曲霉基因组DNA的方法较多,本实验旨在对实验室常用的5种快速提取黑曲霉基因组DNA的方法进行比较,为今后合理选择DNA提取方法及方法的改进提供实验依据。
