摘要
目的:探讨 Jagged1、VEGF 在人脑胶质瘤中的表达。方法收集42例人脑胶质瘤(Ⅰ级9例、Ⅱ级11例、Ⅲ级14例、Ⅳ级8例)及6例正常脑组织标本,采用半定量逆转录聚合酶链式反应技术(RT-PCR)检测肿瘤组和正常组脑组织中 Jagged1的 mRNA 表达水平,用酶联免疫吸附法(ELISA)测定两组患者血清中 VEGF 水平。结果①与正常组相比,肿瘤组 Jagged1 mRNA 表达水平显著升高,差异有统计学意义(P 〈0.05);Ⅲ级与Ⅳ级比较差异无统计学意义(P =0.061),其余两两比较结果差异均具有统计学意义(P 〈0.05)。②与正常组相比,肿瘤组血清 VEGF 表达水平显著升高,差异有统计学意义(P 〈0.05);Ⅰ级与Ⅱ级比较结果无明显差异(P =0.239),Ⅲ级与Ⅳ级比较差异无统计学意义(P =0.089),其余结果两两比较差异均具有统计学意义(P 〈0.05)。③胶质瘤患者Jagged1 mRNA 表达水平与血清 VEGF 浓度呈明显正相关(r =0.986,P =0.014)。结论 Jagged1可能参与了人脑胶质瘤血管的生成,有望作为抗肿瘤治疗的新靶点。
Objective To investigate the expression levels of Jagged1 and VEGF in human brain gliomas. Methods Forty-two cases of human brain gliomas(9 cases in gradeⅠ,11 cases in grade Ⅱ,14 cases in gradeⅢ,8 cases in gradeⅣ)and 6 cases of normal brain tissues were collected after operation. The expression of Jagged1 mRNA was determined by RT-PCR assay. The VEGF level was measured by enzyme linked immunosorbent assay(ELISA). Results ①The expression of Jagged1 mRNA significant-ly increased in human brain gliomas than that in normal brain tissues(P 〈 0. 05),there was no signifi-cant difference between grade Ⅲ and grade Ⅳ in the expression of Jagged1 mRNA( P = 0. 061), and the differences between other grades were significant( P 〈 0. 05). ②The VEGF level significant-ly increased in human brain gliomas than that in normal brain group( P 〈 0. 05);There was no signif-icant difference between grade Ⅰ and grade Ⅱ( P = 0. 239 ),so did grade Ⅲ and grade Ⅳ( P =0. 089),the differences between other grades were significant( P 〈 0. 05). ③The expression of Jag-ged1 mRNA was positively correlated with the level of VEGF( r = 0. 986,P = 0. 014 ). Conclu-sions Jagged1 may take part in human brain gliomas angiogenesis and it may be a novel target for antiangiogenic therapy.
