噻虫嗪大鼠血浆中LC-MS/MS分析方法及其毒代动力学

LC-MS/MS Method for Quantifi cation of Thiamethoxam in Rat Plasma and Its Toxicokinetics Study

摘要 [目的]建立大鼠血浆中噻虫嗪LC-MS/MS分析方法,单次灌胃给予大鼠2种剂量噻虫嗪进行毒代动力学的初步研究。[方法]使用Phenomenex Luna 5μm C18(2)色谱柱,采用ESI离子源,定量离子对为292.0/211.2。采用DAS软件拟合毒代动力学参数。[结果]低高剂量组的毒代动力学参数如下:tmax均为2 h,Cmax分别为71.8、389 mg/L,t1/2z分别为7.84、16.4 h,AUC(0-t)分别为1053、8 661 mg/(L·h)。[结论]该分析方法适用于大鼠血浆中噻虫嗪的测定。噻虫嗪在大鼠体内的毒代动力学过程具有非线性动力学性质。 [Aims] An LC-MS/MS method was developed for the determination of thiamethoxam in rat plasma. Toxicokinetics of thiamethoxam was researched by single oral administration with two doses. [Methods] A Phenomenex Luna 5 pm Cj8(2) column and ESI ion source were used, and quantitative ion pair was 292.0/211.2. The toxicokinetics parameters were fitted using DAS. [Results] The parameters of low and high dose were as follows: t max were all 2 h, C max were 71.8 and 389 mg/L, t 1/2z were 7.84 and 16.4 h, AUC (0-t) were 1053 and 8661 mg/L·h, respectively. [Conclusions] The method was suitable for the analysis of concentrations of thiamethoxam in rat plasma. It was indicated the nonlinear toxicokinetics characteristic of thiamethoxam took place in the studied dose range.
