摘要
目的:通过测定肺癌患者血清中可溶性CD40(sCD40)、可溶性CD40L(sCD40L)、血浆CD62P的含量,探讨血小板源性CD40/CD40L在肺癌外周血中的表达及其临床意义。方法体外分离85例肺癌患者(肺癌组)和25名健康志愿者(对照组)的血清和血浆。用酶联免疫吸附试验(ELISA)法对血清sCD40、sCD40L、血浆CD62P的含量进行测定。结果(1)肺癌组血清sCD40、sCD40L、血浆CD62P含量明显高于健康对照组(P均<0.01)。(2)不同病理类型肺癌患者血清sCD40、sCD40L含量、血浆CD62P含量差异均无统计学意义(P均>0.05)。(3)肺癌患者sCD40、sCD40L、CD62P的含量随临床分期的进展而呈升高趋势。(4)有远处转移肺癌患者sCD40、sCD40L、CD62P的含量明显高于无远处转移患者(P均<0.05)。(5)肺癌患者术后2周时血浆CD62P、血清sCD40L的含量均较术前下降,但血清sCD40含量与术前相比差异无统计学意义。且其CD62P、sCD40L的表达水平成正相关关系(r=0.300,P<0.05)。结论(1)肺癌患者血清sCD40、sCD40L、血浆CD62P含量增加,且表达水平与肺癌的进展程度相关,血小板源性CD40/CD40L系统在肺癌的发病中具有重要作用。(2)检测血小板源性CD40L、CD62P表达水平对研究肺癌的临床分期、预后判断有一定的参考价值。
Objective To investigate the serum concentrations of sCD40, sCD40L and plasma concentrations of CD62P in patients with lung cancer, and to evaluate the clinical signiifcance of CD40/CD40L in lung cancer. Methods Plasma and serum were isolated from 85 patients with lung cancer and 25 normal subjects. The serum concentrations of soluble CD40 (sCD40), soluble CD40 ligand (sCD40L) and the plasma concentrations of CD62P were measured by enzyme linked immunosorbent assay. Results (1)The serum of sCD40, sCD40L and the plasma of CD62P levels in patients with lung cancer was signiifcantly higher than that of normal volunteers. (2)The con-centrations of sCD40, sCD40L and CD62P showed no statistically signiifcant difference among all pathological types of patients with lung cancer (P〉0.05). (3)The levels of sCD40, sCD40L and CD62P elevated with progres-sion of lung cancer. (4)The concentrations of sCD40, sCD40L and CD62P were higher in the patients with metas-tasis than those of without metastasis (P〈0.05). (5)The levels of sCD40L and CD62P in patients with lung cancer decreased when two weeks after surgery, furthermore the levels of sCD40L and CD62P were positively correlated (r=0.300, P〈0.05). The concentration of sCD40 after surgery showed no discrimination with that before surgery. Conclusion (1)These results suggested that there were high levels expression of serum concentrations of sCD40 and sCD40L and plasma concentrations of CD62P in patients with lung cancer. Therefore the levels of them were correlated with the stages of lung cancer. The system of platelet-derived CD40/CD40L might have a potent effect in pathogenesis of lung cancer. (2)Measurement of platelet-derived CD40L and CD62P may therefore provide a tool for monitoring the clinical course of lung cancer.
