高分子量灰树花多糖GFLP的分离纯化及其对免疫细胞的作用 被引量:5

摘要 运用水提、超滤、低浓度醇沉、离子柱层析和凝胶柱层析从灰树花(Grifola frondosa)子实体中分离纯化到一种高分子量多糖GFLP,经高效液相色谱、紫外光谱和离子色谱分析表明GFLP是一种分子量为3037kDa的葡聚糖。体外试验表明:GFLP既能刺激小鼠脾淋巴细胞的增殖,又能显著刺激巨噬细胞RAW264.7分泌NO的活性。 A high molecular polysaccharide(GFLP)was isolated and purified from Grifolafrondosafruit bodies by hot water(100℃)extraction,ultrafiltration,ethanol precipitation(30%),DEAE-Sepharose Fast Flow ion exchange chromatography and Sephacryl S-500 high resolution chromatography.High liquid performance chromatography,UV spectroscopy and ion chromatography analysis indicated that GFLP has a molecular weight of 3037kDa and was composed mainly of glucose.In vitroimmunostimulation experiments revealed that GFLP significantly stimulated the proliferation of mouse spleen lymphocytes and enhanced the release of nitric oxide from RAW264.7macrophages.
