内异康复片对EM大鼠血清及异位内膜细胞中ARO、COX-2、E_2水平的影响

Effect of Neiyi rehabilitation tablets on ARO,COX-2,E_2 levels in serum and ectopic endometrial cells of EM rats

目的:研究内异康复片对子宫内膜异位症(EM)模型大鼠血清中aromatase(ARO)、COX-2、E2的影响,探讨内异康复片治疗EM的机理。方法:体内实验采用手术方法建立EM模型大鼠,大鼠按随机数字表法分为内异康复片高、中、低剂量组、来曲唑组、去势组、模型空白对照组6组;造模成功3周后,给予内异康复片进行干预3周,检测大鼠血清中ARO、COX-2、E2;体外实验从造模成功的EM大鼠上取出大鼠腹腔内异位内膜组织,分离异位内膜细胞进行体外培养,并给予内异康复片干预48 h后,检测细胞培养液中ARO、COX-2、E2的水平。结果:体内外实验结果显示,内异康复片高剂量组、中剂量组可明显降低EM大鼠血清中ARO、COX-2、E2(P<0.05;P<0.01);内异康复片可明显降低异位内膜细胞分泌的ARO、COX-2、E2(P<0.05;P<0.01),体内体外实验结果相似。结论:内异康复片临床治疗EM的机理可能与其抑制异位内膜细胞ARO、COX-2的分泌和降低E2水平相关。

Objective： To study the effects of Nei Yi Kang Fu pill on ARO, Cox-2 and E2 in endometrial rat serum. And to find the mechanism of it to treat EM in clinic. Method： The model of EM was established by operation, the rats were divided randomly into 5 groups： ＂Nei Yi Kang Fu pill＂ high dose group, medium dose group, low dose group and letrozol group, control group. After continuous treatment for 3 w, the level of ARO and Cox-2, E2 in blood serum were detected for each group by ELISA. In the same time,the rat ectopic endometrium were taken out and cultured in vitro, then using Nei Yi Kang Fu pill with different dosages to treat them. After 48 h, the level of ARO, Cox-2, E2 in culture media were detected by ELISA also. Results： The level of E2 , ARO and Cox-2 was significantly different lower compared with control group both of in vivo and in vitro （P 〈0. 05 ;P 〈0. 01 ）. Conclusion： ＂Nei Yi Kang Fu pill＂ can decrease the level of E2 , this maybe correlated with decreasing the levels of ARO and Cox-2, and it can inhibit the growth of the ectopic endometrium in clinic.
