响应面法优化肌苷摇瓶发酵培养基及培养条件 被引量:4

Optimization of Fermentation Medium and Conditions for Inosine Production Using Response Surface Methodology

摘要 采用响应面法对肌苷发酵的培养基及培养条件进行优化。通过Plackeet-Burman设计从肌苷发酵工艺的9个因素中筛选关键因子,结果得出葡萄糖浓度、玉米浆浓度、装液量、接种量4个因子对肌苷产量具有显著的影响。针对4个显著因子进行最陡爬坡试验和Box-Behnken试验,得到参数的最优值和回归模型,并验证结果。最佳工艺参数是:葡萄糖236.87 g/L、玉米浆23.91 g/L、装液量7 mL、接种量6.5%。在最优条件下做验证试验,肌苷产量均值达到8.26 g/L。优化后的肌苷产量较优化前提高了49.9%。 To optimize the medium and conditions for fermentation culture of inosine by response surface analysis.The important parameters were screened by Plackeet-Burman design from 9 factors among inosine shake flask fermentation process and the results showed that inosine yield could be significantly affected by the concentration of glucose,concentration of coin liquid,liquid medium volume and inoculation amount.The path of steepest ascent was used to approach to the biggest region of inosine yield,and the optimum values of the parameters were obtained by Box-Behnken design were as follows: glucose concentration 236.87 g/L,corn liquid concentration 23.91 g/L,liquid medium volume 7 mL and inoculation amount 6.5%.Under such optimum process conditions,the inosine yield reached 8.26 g/L which increased 49.9% than that under the initial conditions.
