摘要
基于色胺具有增强三联吡啶钌(Ru(bpy)32+)电化学发光信号的特性,本文介绍1种毛细管电泳-电化学发光快速分离测定色胺的新方法。重点考察了检测电位、吡啶钌浓度、检测池中缓冲溶液pH值、进样参数等条件对色胺分离检测的影响。结果表明:在最优条件下检测,电位1.15 V,光电倍增管高压800 V,进样条件10kV×10 s,运行高压16 kV,运行缓冲液为pH 6.0 50 mmol/L磷酸盐缓冲溶液。当二盐酸色胺标准样品的浓度在1~8 mmol/L范围,二盐酸色胺的电化学发光强度与其浓度呈良好的线性关系,其线性方程为I=20.649 c+29.839,相关系数r=0.9996(n=8)。检出限(S/N=3)为0.06 mmol/L。将该方法应用于鲳鱼中色胺的分离检测,结果令人满意。该方法具有分析速度快、分离效果好、灵敏度高、重现性好等特点,适用于色胺的快速检测。
A new sensitive method is described for determination of Leucine in beverages by capillary electrophoresis with an electrochemiluminescence detection(CE-ECL) based on tris(2,2'-bipyridyl)ruthenium(II) [Ru(bpy)3 2+].This paper discussed the influence of several experiment parameters such as detection potential,concentration of tris(2,2'-bipyridyl) ruthenium(II),pH of buffer in detection cell,injection parameters(injection voltage and injection time) and running parmeters(separation voltage and pH of running buffer) were explored.The results showed that: under optimal conditions(the detection potential: 1.15 V; photomultiplier tube high voltage: 800 V; injection conditions: 10 kV×10 s; run high voltage: 16 kV; running buffer: pH 6.0 50 mmol/L phosphate-buffer solution) the ECL intensity of trytamine dihydrochloride was linear with concentration of trytamine dihydrochloride in the range of 1~8 mmol/L.The linear equation for I = 20.649c + 29.839,correlation coefficient r=0.9996(n=8) with detection limit(S/N=3) of 0.06 mmol/L.The method used to determinate trytamine in the butterfish samples,the results were satisfied.This method is suitable for the rapid detection of aquatic products,due to its rapid analysis,good separation,high sensitivity and good reproducibility.
