复方水蛭滴眼液对紫外线诱导的人晶状体上皮细胞线粒体跨膜电位的影响 被引量:1

摘要 目的 探讨复方水蛭滴眼液(SZ)防护紫外线(UV)照射诱导的人晶状体上皮细胞(human lens epithelia cell,HLEC)线粒体跨膜电位的影响.方法采用SZ与HLEC共同孵育,再用UV照射HLEC,以吡喏克辛滴眼液(PS)作为阳性对照,观察SZ对紫外线照射的HLEC线粒体跨膜电位的影响.结果 UV照射后细胞活性降低,线粒体膜电位水平下降,SZ可改善UV照射后线粒体跨膜电位下降水平.结论 UV照射可引起HLEC活性下降和线粒体跨膜电位水平下降,SZ对细胞的保护作用是通过提高HLEC活性,升高线粒体跨膜电位来实现的. Objective To investigation the effect of mitochondria trans-membrane potential of compound Shui Zhi eye drops (SZ) on human lens epithelia cells (HLEC) induced by ultraviolet (UV). Methods This work will use human lens epithelia B3 cells. HLEC were cultured and sub cultured in vitro. The cells of SZ group and PS group were incubated with special concentration of SZ and PS respectively. And then,the cells were exposed under UV. After UV irradiation, the irradiated cells were further cultured for 4 hours. (l) The activity of HLEC was detected by methyl thiazolyl tetrazolium (MTT). (2) The mean fluorescence intensity (MFI) of mitochondrial trans membrane potential was detected using flow cytometry. Results The result of MTT test show that UV group can reduce the optical density values (OD) obviously compared to control group(P〈0.01). The OD values of SZ and PS group higher than that of UV group remarkably. The result of MFI of mitoehondrial trans membrane potential show that UV group can reduce the MFI obviously compared to control group(P〈0.01). The MFI values of SZ and PS group is higher than that of UV group remarkably,moreover,the values of SZ group is higher than that PS group(P〈0.01). Conclusion Compound SZ could prevented and treated injury of Human Lens Epithelial Cell induced by UV through raised level of mitochondrial trans membrane potential and elevating activity of HLEC.
