枸杞含药血清预处理对兔视网膜色素上皮细胞光损伤保护的分子机制研究 被引量:2

摘要 目的 观察枸杞含药血清对受损兔视网膜色素上皮细胞中超氧化物歧化酶活性及bc1-2/bax 蛋白表达的影响.方法 常规细胞培养,建立兔视网膜色素上皮细胞光损伤模型,分为正常对照组、模型组、低剂量组(光照前30min+25μl含药血清)和高剂量组(光照前30min+ 25μl含药血清),分别于0、24、48、72h(以光照结束为基点)进行指标的检测.氮蓝四唑法测定兔视网膜色素上皮细胞中超氧化物歧化酶的活性,免疫印迹法检测兔视网膜色素上皮细胞中Bcl-2/Bax蛋白的表达.结果 模型组各时间点兔视网膜色素上皮细胞中SOD活性及Bcl-2/Bax蛋白的表达与正常对照组比较,差异有统计学意义(P<0.05);低剂量组及高剂量组各时间点兔视网膜色素上皮细胞中SOD的活性及Bcl-2/Bax蛋白的表达与模型组比较,差异有统计学意义(P<0.05),且随时间的延长,SOD的活性逐渐增高,Bcl-2蛋白表达逐渐增高,Bax蛋白表达依次降低;高剂量组各时间点兔视网膜色素上皮细胞中SOD的活性及Bcl-2/Bax蛋白的表达与低剂量组比较,差异有统计学意义(P<0.05).结论 枸杞对RPE细胞具有一定的保护作用,这与其能提高受损RPE细胞SOD的活性有关. Objective To observe the superoxide dismutase (SOD) activity and BCL-2/Bax protein expression in rabbit retinal pigment epithelial cells preconditioned with serum containing wolfberry. Methods The light injury model of retinal pigment epithelial (RPE) cells in rabbit was established, and normal control groups, model groups, low-dose group (preconditioned with serum containing 25 μl wolfberry 30 mins before light injury) and high dose group (preconditioned with serum containing 25 μl wolfberry 30 mins before light injury) were contained in this study, and tests were carried 0, 24h, 48h and 72h after lighting. We used blue tetrazole nitrogen to test the SOD activity in retinal pigment epithelial cells in rabbit, and western blot was applied for detection of Bcl-(P〈0. 05), /Bax protein expression. Results SOD activity and Bcl-2/Bax protein expression in model group at all test point were higher than that in normal control groups (P〈0.05), difference were found between low-dose group and high-dose group (P〈0.05), and SOD activity and Bel-2 protein expression in this two groups were increased over time. Conclusion lycium barbarum has play a role in the protection of RPE cells by increasing the SOD activity in damaged RPE cells.
