摘要
目的:通过研究实验探索最佳的醛化剂浓度及醛化时间对红细胞进行醛化处理,以获得保存效期长、凝集效果佳的反定型红细胞试剂。方法:分别以0.5%、0.1%、0.05%的戊二醛对洗涤后的红细胞进行1分钟、3分钟、5分钟、10分钟的醛化,配制成5-10%的醛化反定型红细胞试剂,分别与购买的商品化反定型红细胞试剂,单克隆抗A、抗B正定型试剂进行对比试验,同时在2-8℃冰箱放置进行稳定性试验。结果:用0.1%的戊二醛醛化红细胞可以获得最佳的稳定性与凝集效果的平衡,在12个月的保存试验期内试剂稳定,与商品化的反定型红细胞试剂及正定型的对比实验中,二者的结果完全相符。结论:0.1%的戊二醛醛化反定型红细胞试剂具有良好的稳定性及凝集效果,可以用于ABO血型的反定型,同时用醛化的方法制备稳定而又凝集效果佳红细胞试剂也是一条值得探索的途径。
Objective:Through research experiments to explore the best of hydroformylation concentration and time for hydroformylation processing of red blood cells, to be preserved long preservation period, agglutination good effect against red cell reagents.Methods:0.5%, 0.1%,0.05%, respectively of glutaraldehyde on washing red blood cells, after aldehyde for 1 minute ,3 minutes, 5 minutes, 10 minutes, to prepare 5-10% glutaraldehyde inverse blood grouping red cell reagents, Commercialize anti-stereotyping purchased separately with red cell reagents, monoclonal anti-a, anti-b is in reagents to test, 2-8℃ refrigerators placed at the same time stability testing.Results:0.1% of glutaraldehyde of formaldehyde erythrocytes can balance the best stability and aggregation of results, In the preservation of the 12-month trial period reagents stability, comparison with commodification of inverse blood grouping red cell reagents and positive blood grouping, which exactly matches the results.Conclusions:0.1% inverse blood grouping red cell reagents of hydroformylation of glutaraldehyde with good stability and agglutination effect, can be used in ABO blood group inverse blood grouping, with hydroformylation method in preparation of stable and agglutination good effect of red blood cell reagent is also a worth exploring ways.
