摘要
目的探讨全反式维甲酸(ATRA)对卵巢上皮性腺癌细胞株(COC2)增殖的抑制作用。方法用不同浓度的ATRA处理体外培养的COC2,采用直接细胞计数法计算一定时期内的细胞密度情况;利用倒置显微镜和电镜观察细胞形态学改变及细胞结构变化。结果 ATRA能够抑制COC2增殖,在1~20μmol/L之间,具有浓度依赖性,在30μmol/L时几乎未见存活细胞;倒置显微镜和电镜观察到使用ATRA处理后,细胞形态及结构受到破坏。结论在一定浓度范围内,ATRA对卵巢上皮性腺癌细胞株(COC2)有抑制作用。
Objective To investigate the effects of all-trans retinoic acid ( ATRA ) on inhibitory cell proliferation and inducting differentiation in human ovarian cancer cell line COC2. Methods All the ovarian cancer cells were treated respectively at different ATRA concentrations and were cultured in the same conditions. The cells were evaluated separately by direct cell counts, and cell morphological changes were observed by inverted microscope, and cell ultrastructures were observed by electronic microscope. Results Cell growth inhibition occurred in COC2 cell lines in a dose dependent manner within a certain concentration ranged in ATRA 1-20 μmol/L and no ceils were observed in concentration of 30μmol/L. Cell growth overlapping was observed by inverted microscope and electronic microscope after the cells were treated by ATRA. Conclusions It is suggested that ATRA was effective agent on inducing differentiation and promoting apoptosis in human ovarian cancer cell line ( COC2 ) in a certain concentration range.
