糖尿病和结直肠癌双重疾病动物模型的建立和应用 被引量:1

Establishment and application of animal model with double diseases of diabetes and colorectal cancer xenongraft

摘要 目的建立肠癌移植瘤合并2型糖尿病(T2DM)小鼠动物模型,检测移植瘤组织中血管内皮生长因子(VEGF)的表达。方法 48只ICR小鼠随机分为肠癌糖尿病组(A组,14只)、糖尿病组(B组,10只)、肠癌组(C组,14只)和空白对照组(D组,10只)。A组、C组小鼠右腋皮下移植小鼠结肠癌细胞株CT26,建立肠癌移植瘤合并T2DM双重动物模型。观察移植瘤体积的变化,免疫组化方法检测移植瘤VEGF的表达。结果 A组和C组小鼠在接种后第7天肿瘤组织体积均逐渐增大,A组瘤体体积增大更为明显。A组移植瘤组织VEGF表达高于C组[(71.6±8.7)%vs.(42.0±10.4)%](P<0.05)。结论成功建立了糖尿病和结直肠癌双重小鼠动物模型。糖尿病可能为促进肠癌生长的因素之一。 Objective To establish a mouse model with double diseases of diabetes mellitus (T2DM) and coloreetal cancer xenongraft and detect the expression of vascular endothelial growth factor(VEGF). Methods Forty-eight ICR mice were randomized into 4 groups of A(with double diseases of T2DM and colorectal cancer xenongraft, 14 mice), B(with T2DM, 10 mice), C (with coloreetal cancer xenongraft, 14 mice) and D(btank control, 10 mice). The coloretcal cancer CT26 was inoculated in subcutaneous part of right forelimb armpit in the mice of group A and group C. The changes in the volume and size of the xenongrafts were observed and VEGF expression in the xenongrafts were determined by immunohistochemical staining. Results The mouse model with double diseases of T2DM and coloreetal cancer xenongraft was established successfully. The growth and volume of xenongrafts developed more in group A than those in group C. The expression of VEGF in the xenongrafts was significantly higher in group A than that in group C[-(71.6±8. 7)% vs. (42.0±10. 4)%](P〈0. 05). Conclusion The mouse model with double diseases of T2DM and colorectal cancer xenongraft has been established successfully. T2DM may be one of the factors for promoting the growth of colorectal cancer.
