摘要
目的:研究CyPA、CD147、MMP-2、MMP-9在环孢素诱导大鼠牙龈增生模型中的表达,探讨环孢素诱导牙龈增生的发病机制。方法:将5周雄性SD大鼠48只随机分为2组(n=24)。实验组大鼠给予环孢素注射液皮下注射,对照组给予等量橄榄油皮下注射,4周后全部处死,取牙龈组织。用免疫组化和Western blot检测CyPA、CD147、MMP-2、MMP-9的定位和定量。结果:免疫组化结果显示,实验组较对照组CyPA、MMP-2、MMP-9阳性细胞平均吸光度值降低(P=0.000);实验组与对照组CD147的表达无明显差异(P>0.05)。Western blot显示实验组较对照组CyPA、MMP-2、MMP-9蛋白的表达量减少;CD147的表达无明显差异。结论:CsA可能通过下调CyPA的表达,减少MMPs的分泌,引起牙龈胶原代谢失衡,导致牙龈增生。
Objective: To investigate the expression of CyPA, CD147, MMP-2 and MMP-9 in eyclosporin-induced gingival overgrowth of rats. Methods: 48 male SD rats were divided into 2 groups randomly ( n = 24). Cyclosporin and olive oil were subcutaneously injected into the mice in experimental and control groups respectively. Immunohistochemistry and western blot was used to detect the expression of CyPA, CD147, MMP-2 and MMP-9 in gingival'tissue. Results: Western blot and immunohistochemistry revealed CyPA, MMP-2, MMP-9 expressions were decreased in the rat gingival tissue in experimental group compared to that in the control group (P = 0. 000). CD147 expression did not show statistic difference (P 〉 0.05 ). Conclusion: Cyclosporine may down-regulate the expression of CyPA, decrease the secretion of MMPs, induce the imbalance of collagen metabolism and result in overgrowth of gingiva.
