摘要
首次研究了两面针提取物在大鼠胆汁中的代谢。大鼠灌胃后,收集胆汁,用HPLC-DAD/ESI-Q-TOF-MS分析,经与空白样本及药材体外分析结果比对,寻找可能的药物原型及代谢产物。本实验一共鉴定了44个化合物,还发现多数成分的代谢方式为还原、甲基化、去甲基化、氧化,为两面针的后续药理学研究奠定了一定的基础。
The compounds in bile after oral administration of Zanthoxylum nitidum were identified by HPLC - DAD/ ESI - Q - TOF - MS. After garage feeding in rats of the plant extract, bile samples were collected and analyzed by high - performance liquid chromatography with diode array detection coupled to quadrupole time - of - flight mass spectrometry ( HPLC - DAD/ESI - Q - TOF - MS). A total of 44 compounds were identified or tentatively characterized on the basis of their accurate prorogated molecular masses and fragmentation behaviors. The results suggested that reduction, methylation, oxidation glucuronidation and sulfation were the main metabolic pathways of alkaloids. It was the first report on the metabolic profiling of alkaloids from Zanthoxylum nitidum and the method developed could be applied for the structural elucidation of metabolites of other compounds.
