两种前列腺癌细胞株中E钙粘素和波形蛋白的表达意义

摘要 目的探讨前列腺癌细胞骨转移的机制及LNCaP和IA8两种细胞株中的E钙粘素以及波形蛋白的表达意义,对前列腺癌治疗的分期及药物使用进行指导。方法对前列腺癌LNCaP和IA8两种细胞株中的E钙粘素以及波形蛋白的表达情况使用Western印迹法进行鉴定,分析其不同的转移潜能以及在前列腺癌的转移过程中的作用机制。结果 E钙粘素在LNCaP中存在高表达,在IA8中却缺失,而波形蛋白却在IA8中存在高表达,在LNCaP中缺失,可见E钙粘素和波形蛋白在LNCaP和IA8细胞表达中差异有统计学意义(P<0.01),而β-肌动蛋白在二者的表达中差异无统计学意义(P>0.05),但二者的E钙粘素/β-肌动蛋白及波动蛋白/β-肌动蛋白差异有显著统计学意义(P<0.01)。结论前列腺癌患者的LNCaP和IA8细胞株中确实存在着E钙粘素和波形蛋白表达差异,说明E钙粘素和波形蛋白在前列腺癌的骨转移过程中发挥着促进或者抑制作用,这对探讨前列腺癌细胞骨转移的机制和治疗并最终对抗前列腺癌有重要意义。 Objective To explore the mechanism of prostate cancer bone metastases and LNCaP and IA8 two cell lines of E calcium element and the expression of vimentin and to guide the treatment of prostate cancer staging and drug using. Methods The prostate cancer of the two kinds of cell lines of E calcium element and the expression of vimentin using Western imprinting method for appraisal,analyse of the different transfer potential and mechanism of action in the transfer process of prostate cancer. Results E stick calcium element in LNCaP high expression,but lack in IA8,while vimentin IA8 existed in the high expression of calcium loss in LNCaP,visible E glue and vimentin in LNCaP and IA8 cells expressed a significant difference(P &lt; 0. 01),while β-actin in both the expression had no significant difference(P &gt; 0. 05),but both calcium E stick element /β-actin and volatility protein /β-actin was significant difference(P &lt; 0. 01). Conclusion The prostate cancer patients LNCaP and IA8 cell lines does exist in E calcium stick and waveform protein expression differences,E calcium sticky grain and vimentin in the process of the bone metastasis of prostate cancer,play an important role to promote or inhibit it to explore the mechanisms of prostate cancer bone metastasis and eventually has important significance in the fight against prostate cancer.
