摘要
目的观察辛伐他汀对胃癌的致凋亡活性,并探讨其作用机制。方法 CCK-8细胞增殖实验检测辛伐他汀对胃癌SGC-7901的生长抑制活性,细胞凋亡的发生使用流式细胞仪检测,CCK-8细胞增殖实验检测巨核细胞增殖能力的变化,Western blotting检测生存素(Survivin)的表达。结果辛伐他汀对SGC-7901胃癌细胞IC50值为(12.72±1.04)μmol/L。0、20、40μmol/L的辛伐他汀作用胃癌SGC-7901细胞48 h后,SGC-7901细胞凋亡率分别为(3.74±1.26)%、(33.28±6.53)%和(52.15±11.62)%,各组之间差异具有统计学意义(P<0.01)。20μmol/L和40μmol/L的辛伐他汀作用胃癌SGC-7901细胞48 h后,SGC-7901细胞Survivin蛋白的表达显著降低。结论辛伐他汀可以显著抑制胃癌细胞的生长,致胃癌细胞凋亡,下调Survivin蛋白表达可能是其抗胃癌的主要作用机制。
【Objectives】 To study the apoptotic activity of Simvastatin against gastric cancer cell and explore its action mechanism. 【Methods】 CCK-8 assay was used to detect the inhibited activity of Simvastatin against gastric cancer SGC-7901 cells. The apoptosis was determined by flow cytometry analysis. The protein expression of Survivin was detected by Western blot analysis. 【Results】 The IC50 values of Simvastatin against gastric cancer SGC-7901 cells was 12.72 ± 1.04 μmol/L. After treatment with 0, 20, and 40 μmol/L Simvastatin for 48 h, the percent of apoptosis was elevated from 3.74 ± 1.26 to 33.28 ± 6.53 and 52.15 ± 11.62%. There was significant difference among the different treatment(P〈0.01). Treated with 0, 20, and 40 μmol/L Simvastatin for 48 h, the protein expression of Survivin was down-regulated. 【Conclusion】 Simvastatin showed effective activity to inhibit the prolifecation and induce apoptosis of gastric cancer cells. Down-regulation of Survivin maybe its action mechanism.
