摘要 目的:观察人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)对Aβ诱导的人胚胎大脑皮质神经元细胞活力的影响。方法:人胚胎大脑皮质神经元原代培养后,以hTERT重组腺病毒转染神经元,用Aβ25-35诱导神经元的凋亡,72 h后用MTT检测各组细胞数量和活力,并计算其保护率。结果:hTERT重组腺病毒转染后72 h进行Aβ干预,此时hTERT对神经元的保护力为29.8%,活细胞的数量及细胞活力(0.310±0.033)明显高于非转染组(0.237±0.081),P=0.002,差异有统计学意义。结论:Aβ干预后的神经元,其活力明显下降,而hTERT重组腺病毒转染可以促进神经元存活,对Aβ干预的人胚胎大脑皮质神经元具有保护作用。 Objective:To observe the effects of hTERT on human embryonic cerebral cortex neuron after Aβ invention. Methods:Primary cerebral cortex neurons of human embryo that was cultured were transfected with hTERT. After the neu-rons were dealt with Aβ25-35 ,the activity of neurons was measured by MTT. Result:After 72 hours transfection with pSU-CMV-hTERT,the neurons were intervented of Aβ,the neurons could resist the toxic effects of Aβ effeciently,the protec-tive index is about 29. 8%,and the activity of neurons(0. 310 ± 0. 033)was significanly higher than that without treat-ment(0. 237 ± 0. 081)detected by the MTT,P=0. 002. Conclusion:The vitality of the neurons intervented with Aβob-viously was decreased. however,the hTERT transfection could promote the survival of neurons,and show the protective effects to neurons.
